Research Background
Semaglutide is a synthetic analog of glucagon-like peptide-1 (GLP-1) that binds the GLP-1 receptor (GLP-1R), a class B G protein-coupled receptor. Receptor-signaling studies describe GLP-1R activation as coupling primarily to the Gαs/adenylate cyclase pathway, raising intracellular cAMP in a glucose-dependent manner in pancreatic beta-cell models (see mechanism review, PMC, U.S. National Library of Medicine). Structural modification of the peptide backbone — including a fatty-acid side chain for albumin binding — is described in the analog-design literature as altering the pharmacokinetic profile of receptor engagement relative to native GLP-1.
