Research Background

Tirzepatide is a synthetic peptide that binds and activates both the glucose-dependent insulinotropic polypeptide receptor (GIPR) and the glucagon-like peptide-1 receptor (GLP-1R). In vitro receptor-pharmacology studies characterize tirzepatide as an imbalanced, biased dual agonist: its activity at GIPR closely resembles native GIP signaling, while its activity at GLP-1R is biased relative to native GLP-1, with reduced recruitment of beta-arrestin relative to cAMP signaling (Willard et al., JCI Insight, 2020). This receptor-binding profile is what distinguishes tirzepatide from single-receptor GLP-1 agonists in the published pharmacology literature.
